KUMC Transgenic and Gene-Targeting Institutional Facility

Overview of Services

      

 

The Transgenic and Gene-Targeting Facility is an institutional core research support facility providing gene-modified mice and pluripotent stem cells to investigators at KUMC and the surrounding Kansas City research community. In addition, the Transgenic Facility is available for consultation and provision of special services.  The Facility services include:

 Embryo Manipulation Services

  • Modification of zygotes to generate transgenic and gene-modified mice 
    • Inludes hemizygous transgenics, deletions, point mutations, knockins and floxed alleles
    • Includes transgenes and genome-editing elements (CRISPR/Cas9, TALENs and ZFNs)
    • Includes microinjection, electroporation and in situ electroporation
  • Injection of blastocyst stage embryos with embryonic stem cells to generate chimeric mice
  • Sperm cryopreservation
  • Embryo cryopreservation
  • In vitro fertilization
  • Embryo transfer rederivation

Cell Culture Services

  • Mouse embryonic fibroblasts (MEFs)
  • Mouse ES cell targeting
  • Mouse ES cell chromosome count (karyotyping)
  • Derivation of human primary fibroblasts from skin biopsy
  • Human iPS cell generation via reprogramming
  • Gene targeting in human PSC
  • Differentiation of human PSC
  • Human PSC chromosome count (karyotyping)
  • Mycoplasma testing
  • Teratoma formation assay

CRISPR Design and Molecular Biology Services

  • Primer optimization
  • Development of CRISPR reagents (includes design, crRNA, PCR optimization, and RNP reagents)
  • Validation of CRISPR reagents in mouse embryos
  • Validation of CRISPR reagents in mouse ES cells
  • Genotyping
  • Genotyping 96-well plate of pluripotent stem cells
  • Stock development from founders to F1 heterozygotes
